The first thing to check is the RF Power slider:
Also check TX Band Settings to be sure that PTT Inhibit is not set for the band that you are on.
There are quite a few other things to check, so if this is not the problem, please post a screen shot of SmartSDR to make it a bit easier to spot any issues.

Everything looks OK… I do see that you are transmitting on ANT 2. Is it possible that there is no antenna connected to that port?

Also I see that FDX is enabled. I don't think that is in the way, but you might want to turn it off.

i will try turning off fdx in the morning as i have my 6400 conected tonight0

HMMMM… looking at the screenshot, I also note that this is on 6m? Is the output any different on other HF bands?
Also, it looks like the TUNE button is being activated. Is it possible that the TUNE output is set very low? or zero?
Also, the ATU appears to be bypassed. Is the antenna a good match? Is there another antenna tuner in the output?

One more thought … what third party s/w are you running ? Some third party software has extensive control capability through the Flex API. You might check or even just shut down any such s/w to test.

Full Duplex being on or off would not have any impact on this scenario.
Let us know how you make out.

in the screenshot, the TUNE power looks to be set at default 10 watts. Set it in Settings/TX Band Settungs
