From the info I gathered at Hamvention, the 6400 is using an updated digitizer, has a 3rd order preselector, has some added connectivity at the rear, the option for the front panel controls, has optional GPSDO module, modular design for easy repair and possible hardware upgrades.

That is a good summary. In addition, there is a MARS/SHARES filter module that will allow for out of band transmit that was not possible on the 6300.
And you have to purchase the "M" model to get the front panel controls; that is not an option.1 -
I am not a technical person. Could you explain in simple language what a digitizer is and what a 3rd order preselector does?

The ADC (digitizer) takes an analog RF signal and converts it into a binary bit stream (ones and zeros) so it can be processed by digital signal processing software.
A preselector is a band pass filter. It allows signal within a band to pass through the filter and rejecting signals outside of the filter's passband. By rejecting strong out of band signals, it minimizes potential interference within the band of interest.
3rd order is a description of how must signal rejection the filter is capable of providing. The higher the order, the greater the amount of rejection.
